The fire at the Jazan General Hospital, in the south of the country, began on the first floor in the intensive care unit and the maternity ward.
The injured had been transferred to private and general hospitals in the area, it added.
Photographs published on Twitter showed intense black smoke and blaze which appeared to have severely damaged the interior of the three-storey hospital.
Others show the scale of damage inside the building and a number of survivors being treated nearby.
An investigation is under way.
The blaze has prompted criticism on social media about the "disastrous situation" of hospitals in the region, with calls for the oil-rich kingdom to put more investment into infrastructure.
